The majority of in vivo microdialysis sampling experiments are performed with the use of a single probe implanted into a tissue. When sampling from heterogeneous tissues, such as the stomach, differences between the tissue layers cannot be determined by sampling with a single probe. Currently, microdialysis sampling experiments are performed with a probe implanted into the submucosa to characterize compounds from both the mucosa and submucosa layers. Microdialysis sampling in each layer would be a better representation of sampling from the stomach. The goals of my research were to develop multiple probe microdialysis sampling in the different tissue layers of the stomach. Methods of probe implantation simultaneously into the rat stomach lumen, mucosa, submucosa and also in the blood were developed. To determine the significance of multiple probe sampling, this approach was used to monitor drug absorption in the stomach. Concentration-time curves and pharmacokinetics analyses were performed to determine differences in drug concentration from each probe location. To further expand the use of multiple probe microdialysis sampling in the stomach, a similar approach was developed to directly compare different tissue types (healthy and ulcerated tissue) in the same rat stomach. After chemical ulcer induction, methods of probe implantation simultaneously in the stomach lumen, submucosa of both normal and ulcerated tissue and in blood were developed. This approach was then utilized to monitor drug absorption through the stomach to determine absorption differences in ulcerated and normal tissue.
Personal Interests:
My husband, Daniel, and I enjoy spending time with our families, hanging out with friends on our house deck, watching KU basketball and watching movies. I am originally from the Chicago area, which would explain my love for the Chicago Cubs and Bears. In terms of playing sports, since I was raised in a family of bowlers, it was inevitable that I would become a bowler too. I have a little boston terrier named Danie, who is such a sweetheart and a fabulous snuggle buddy! I love all of my family and friends near and far. Mom, I love you and miss you everyday.
